ABSTRACT Sequences of the coat protein (CP) and 3'-end nontranslated region (3'NTR) of 13 isolates and the helper component proteinase (HC) of nine isolates of potato A potyvirus (PVA) were determined and compared with the eight previously determined PVA CP and 3'NTR sequences and one HC sequence. CP amino acid (aa), 3'NTR nucleotide, and HC aa sequence identities were 92.9, 93.4, and 94.8%, respectively. Sequence data, serological tests, and the necrotic local lesions induced in the leaves of the potato hybrid 'A6' confirmed that tamarillo mosaic virus is a strain of PVA. The aa substitutions A6T and G7S in the CP N-terminus were correlated with loss of aphid transmissibility. Development of necrotic lesions or nonnecrotic symptoms in the systemically infected leaves or lack of systemic spread in potato cv. King Edward were used to place the PVA isolates into four strain groups, but this grouping was not correlated with any differences in CP, HC, or 3'NTR. Recognition of CP by three monoclonal antibodies was used to place the PVA isolates into three groups different from the four groups above. The epitopes of two mono-clonal antibodies were mapped by site-directed mutagenesis to the same lysine residue at the CP aa 34.  相似文献

The adhesion of a spray droplet upon initial contact with a leaf surface is extremely important to spray efficacy and is dependent on dynamic interactions between droplets (formulation, size, velocity) and leaf (micro‐topography, surface chemistry, veininess, hairiness and orientation). A ‘universal’ spray droplet adhesion model has previously been developed, using 50% aqueous acetone contact angles as a measure of leaf surface properties; this model satisfactorily predicts initial adhesion over a range of formulation surface tensions, droplet sizes and velocities. However, it failed to fit data from hairier leaves. This study investigates initial spray droplet adhesion on hairy leaves. Two categories of hairy leaves were identified by how the droplets penetrate the leaf hairs, Wenzel (hairy) and Cassie–Baxter (super hairy). For the Wenzel‐type, a simple constant accounted for the increased droplet shatter caused by the hairs. For the Cassie–Baxter‐type, a cushioning factor was introduced to account for the absorption of kinetic energy at impact by the hair mat. The cushioning factor was estimated by measuring the relative height of the hair mat. By including these two parameters, the new model successfully predicted the mean adhesion of non‐hairy, hairy and super‐hairy plants (R2 = 0.96). This model and the underlying principles determining hairy leaf adhesion developed in this article will help develop spray formulations effective at targeting hairy‐leaved weed and crop species.  相似文献

BACKGROUND: Flea allergy dermatitis (FAD) is a common skin disease in dogs and can be induced experimentally. It often coexists with other allergic conditions. So far no studies have investigated the quantitative production of cytokine mRNA in skin biopsies and peripheral blood mononuclear cells (PBMC) in flea allergic dogs. OBJECTIVE: The aim of our study was to improve the understanding of the immunopathogenesis of allergic dermatitis as a response to fleabites. MATERIAL AND METHODS: Allergic and non-allergic dogs were exposed to fleas. Before and after 4 days of flea exposure mRNA was isolated from biopsies and PBMC. Production of chymase, tryptase, IL-4, IL-5, IL-13, TNF-alpha and IFN-gamma mRNA was measured by real-time RT-PCR. The inflammatory infiltrate in the skin was scored semi-quantitatively. The number of eosinophils, mast cells (MC) and IgE+ cells/mm2 was evaluated to complete the picture. RESULTS: FAD was associated with a higher number of MC before flea exposure and with a significant increase of eosinophils after flea exposure as compared to non-allergic dogs. The number of IgE+ cells was higher in allergic dogs before and after flea exposure. In allergic dogs mRNA for most cytokines and proteases tested was higher before flea exposure than after flea exposure. After exposure to fleas an increased mRNA production was only observed in non-allergic dogs. In vitro stimulation with flea antigen resulted in a decreased expression of most cytokines in allergic dogs before flea exposure. In contrast, in PBMC, only increased levels of IL-4 and IL-5 mRNA were observed in allergic dogs before flea exposure. However, after flea exposure and additional stimulation with flea antigen the production of mRNA for all cytokines tested was significantly increased in allergic dogs. CONCLUSION: We demonstrated that the response in biopsies and PBMC is different and that FAD is associated with a TH2 response.  相似文献

Four breviaristatum (short awned and semi-dwarf) barley mutants; ari-e.1, ari-e.119, ari-e.156 and ari-e.228 were compared with other semi-dwarf mutants; Golden Promise, Alf, Pallas and Diamant along with their non-mutant parents; Bonus, Foma, Maythorpe, Bomi and Valticky, for response to salt stress. Plants were exposed to hydroponic salt treatments (NaCl at 25 and 175 mol m-3) for 4 weeks, after which response was measured in terms of shoot dry weight, sodium content and δ13C. In general ari-e mutants and Golden Promise had significantly lower Na+ contents than the other mutants. They also had significantly more negative δ13C values than the other lines in stressed (175 mol m-3 NaCl) conditions. There was a positive correlation (r = 0.71, p < 0.01) between shoot Na+ and δ13C values so that δ13C became less negative with increasing Na+ content. Shoot dry weights were compared to shoot Na+ and δ13C values. The ari-e and Golden Promise mutants showed less reduction in dry matter production in salt stress relative to the control treatment than all the other lines. The data suggest that ari-e mutants and Golden Promise are better adapted to salt stressed environments than the other lines examined. Tests for gibberellic acid sensitivity revealed that ari-e mutants and Golden Promise responded weakly to GA3, while other dwarf mutants Pallas, Diamant and Alf along with their parents Bonus, Foma, Maythorpe, Valticky and Bomi were highly sensitive. Our results support previous findings that ari-e mutants and the GPert mutant are allelic. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The main objective of this work is to demonstrate the expression of salt tolerance genes in a wheat-Agropyron amphiploid. Salt tolerance tests were carried out on wheat varieties, ‘Chinese Spring,’ and ‘Glenn-son 81’ the amphiploid between ‘Chinese Spring’ and A. junceum, A. junceum and amphiploid × wheat hybrids, Apart from germination in petridishes all other tests were carried out on plants grown in saline hydroculture tanks. Fresh weight measurements are given for stressed and non-stressed plants as well as measurements of harvest ripe plants. The utility of A. junceum as a source of salt tolerance genes for wheat is discussed.  相似文献

The contents of moisture, protein, ash, ascorbic acid, glucose, fructose, total sugars, and total and insoluble fiber were determined in cultivars of bananas (Gran Enana and Peque?a Enana) harvested in Tenerife and in bananas (Gran Enana) from Ecuador. The chemical compositions in the bananas from Tenerife and from Ecuador were clearly different. The cultivar did not influence the chemical composition, except for insoluble fiber content. Variations of the chemical composition were observed in the bananas from Tenerife according to cultivation method (greenhouse and outdoors), farming style (conventional and organic), and region of production (north and south). A highly significant (r = 0.995) correlation between glucose and fructose was observed. Correlations of ash and protein contents tend to separate the banana samples according to origin. A higher content of protein, ash, and ascorbic acid was observed as the length of the banana decreased. Applying factor analysis, the bananas from Ecuador were well separated from the bananas produced in Tenerife. An almost total differentiation (91.7%) between bananas from Tenerife and bananas from Ecuador was obtained by selecting protein, ash, and ascorbic acid content and applying stepwise discriminant analysis. By selecting the bananas Peque?a Enana and using discriminant analysis, a clear separation of the samples according to the region of production and farming style was observed.  相似文献

The effects of the interactions between higher plants and microorganisms and the addition of selected microorganisms on aggregation of sand from an embryo dune were examined. Addition of microorganisms to sand increased both plant growth and the amount of aggregation. Roots alone had little effect on aggregation but in association with microorganisms there was a noticeable increase. The number of microorganisms isolated from aggregates in four different treatments varied each month over the course of the experiment. Aggregates from a treatment receiving Penicillium spores were colonized by the greatest number of microorganisms, and the least colonized aggregates were from a treatment where the sand was initially sterile.  相似文献

Starch content of potato tubers in relation to K nutrition and tuber growth In a pot experiment with potato (cv Saturna) K content in tubers increased from 1.0 to 2.6 % of tuber-DM due to increasing rates of K fertilizer. Simultaneously starch in tuber fresh matter decreased from 21 to 15 %. Highest starch content in tuber dry matter was obtained in that K treatment which gave the highest yield in dry tubers. Measuring starch accumulation during tuber growth revealed that already in the flowering stage differences exist in the starch contents of the tuber dry matter between the lowest and highest K treatments, namely 69 % for K1 as compared with 64 % for K5. The rate of starch accumulation between flowering and final harvest, however, was similar in all K treatments. This suggests that K supply affects the storage tissue at an early stage of tuber development probably via different water relations of tubers.  相似文献

Holstein steer calves (101 to 350 kg BW) consumed bermudagrass hay ad libitum without or with supplemental ground corn up to 1.0% of BW. As BW increased, total DMI increased quadratically (-2.459 + .05448 [BW]-.000073 [BW2] + .540 [corn DMI]; R2 = .83, sy.x = .655). Each kilogram of corn DM decreased bermudagrass DMI by .46 kg. Total digestible OM intake (kg) increased with BW and corn supplementation (.314 + .0127 [BW] + .441 [corn OM intake]; R2 = .79, sy.x = .444). Feed intake level accounted for approximately 2.5 times more variability in total digestible OM intake than digestion did. Corn supplementation decreased digestion of bermudagrass NDF (62.50 - 8.468 [corn DMI, % BW]; R2 = .13, sy.x = 8.121), with a similar decrease across BW. Increasing bermudagrass DMI (% of BW) decreased bermudagrass NDF digestion slightly, but variation accounted for was only 33% of that attributable to corn DMI (% of BW). Concentrations of common fiber fractions (NDF, ADF, cellulose, hemicellulose, and ADL) in bermudagrass explained very little variation in feed intake and digestion, indicating considerable influence of other factors. Bermudagrass intake and digestion were not related, and no substantial interactions were observed among steer BW, corn level, and bermudagrass composition.  相似文献
